Fill/finish operations represent perhaps the last frontier of significant risk in manufacturing injectable medicines. Closed systems in compounding and formulation, and sterile filtration at the point of filling using validated, ready-to-use components with engineered sterile connectors virtually eliminate threats to product purity up to this stage. Risk remains in the momentary exposure of components and product during filling and before closing. Barrier technology in the form of restricted access barrier systems (RABS) and isolators have been utilized for decades, yet legacy facilities continue to operate with people around open processes.
